You will set up a 3 ml enzyme assay in a plastic cuvette. Imagine you have available to you a
0.5 M sodium phosphate buffer at pH 6.5, and a 10 mM substrate solution, an enzyme solution of
unknown concentration, and ddH2O.
The assay you¥ setting up must have of 100 ?l of the enzyme solution, a final substrate
concentration 3 mM, and a final phosphate concentration of 100 mM.
Calculate the volume of water, phosphate buffer, substrate stock solution and enzyme (given
above) that should be added to give a 3 ml reaction.
